Dietary and pharmacological treatment of diet-induced obese mice – impact on mitochondrial function.

To determine the impact of (i) dietary supplementation with Calanus oil (a novel marine oil extracted from the marine copepod Calanus finmarchicus) and (ii) infusion of exenatide (a GLP-1 anaog) on mitochondrial respiration in various organs of obese mice, including adipose tissue, liver, heart and skeletal muscle.

Six weeks old female mice (C57bl/6J) received a high-fat diet (HFD, 45% energy from fat) for an initial period of 12 weeks. Thereafter they were separated in 3 subgroups which were fed with either HFD, HFD supplemented with 2% Calanus oil (HFD+Cal), or HFD combined with administration (via mini-osmotic pumps) of 10 µg/kg/day exenatide (HFD+Ex). Mitochondrial respiration was measured in homogenates of liver, heart, and intra-abdominal white adipose tissue (WAT), as well as in permeabilized fibers of skeletal muscle.

Preliminary data indicate that respiration was reduced in liver, WAT and skeletal muscle of (HFD+ Cal) mice compared to the non-supplemented HFD group, while it was increased in cardiac tissue. Exenatide also seemed to reduce respiration in skeletal muscle and heart, while increasing respiration in liver and WAT. These data will be presented and discussed during the training school.

Dietary supplementation with Calanus oil, as well as administration of exenatide, impacts on mitochondrial respiration in major organs of mice fed a high-fat diet. The data need, however, further interpretation and statistical evaluation.
