Abstract: Course in Mitochondrial Medicine, Bologna, IT; Course in Mitochondrial Medicine 2014.

Course description: Mitochondrial Medicine is a relevant, steadily expanding area of medicine. The complexity of the metabolic pathways associated with mitochondrial energy homeostasis and the peculiar genetics dictating mitochondrial inheritance in humans, account for the extraordinarily wide spectrum of clinical presentations and genetic abnormalities associated with mitochondrial disorders. This Course on Mitochondrial Medicine intends to offer to European biochemists and biologists, geneticists and physicians the basic and clinically-oriented information on this frontline scientific field, through an articulate program organized in morning plenary lectures and afternoon Meet Researchers presentations.

In WP1: investigating the relationship between mitochondrial genetics and metabolism and tumor formation/progression:

  • Targeting complex I (cI) as an anticancer strategy
  • Metabolic targeting of solid cancers

In WP2: elucidating the molecular mechanisms underlying segregation, selection and (epi)genetic control of mitochondrial DNA mutations, in order to formulate therapeutic strategies for mitochondrial disorders:

  • Preventing germline transmission of pathogenic mtDNA mutations
  • Molecular basis of the spontaneous recovery in infantile reversible cytochrome c oxidase deficiency myopathy
  • MtDNA deletions accumulation and their clonal expansion in tumorigenesis and neurodegeneration
  • Manipulating segregation of human mtDNA

In WP3 : unveiling the relationship between mutations and their functional effect on mitochondrial energetic efficiency, focusing on the relevance for human pathologies for which in vitro and in vivo models may be created:

  • Structure, function and assembly defects of cI: comprehensive identification and characterization of cI chaperones
  • Cofactor deficiency in mitochondrial diseases
  • Determine OXPHOS organization and identify the factors that drive and modulate interaction between respiratory complexes
  • Respiratory complexes assembly and mitochondrial lipid metabolism

In WP4 : implementing selection of potential drugs that may enter pre-clinical and clinical trials for mitochondrial disorders. This WP intends to bridge basic and translational to clinical research, providing an applicative twist to the MEET bench-to-bed approach:

  • Optimizing techniques for high-content drug screening in vitro
  • From compound selection, modification and pharmacology to therapy trials and toxicology in animal models
  • Therapeutic approaches in animal models of mitochondrial disease
