We aim to summarize and analyze, from a technical perspective, data collected on high-resolution respirometry in permeabilized skinned muscle fibers during the last ten years. Our work is mainly performed in mouse models where physiological interventions are performed to evaluate metabolic adaptations. Exercise training, dietary interventions to induce obesity-related type 2 diabetes or genetically modified mouse models are the main experimental approaches. Our overall interest is to assess metabolic plasticity after different physiological and pathophysiological challenges. In this occasion we are using most of the data recorded along these years to open a discussion about the strength of the technique and also to identify weak points where more work should be done to improve the consistency of the assays.
