Description
Salicylhydroxamic acid (SHAM; synonym: 2-Hydroxybenzohydroxamic acid N,2-Dihydroxybenzamide) is an inhibitor of the alternative oxidase (AOX). When AOX is blocked by SHAM, electrons are forced through the CIII-cytochrome c oxidase pathway, allowing observation of the operation of the CIII-CIV pathway without AOX activity.
Abbreviation: SHAM

Application in HRR
- SHAM: Salicylhydroxamic acid (99%; HOC6H4CONHOH); Sigma S 607, 5 g, store at RT; M = 153.14 gΒ·mol-1.
- SHAM is reported to be easily soluble in EtOH. So EtOH could be considered as a solvent, too. However, in Oroboros-lab SHAM in EtOH has not been tested yet.
- Caution: Irritant!
- Preparation of 250 mM stock solution (dissolved in DMSO):
- Weigh 76.57 mg of SHAM.
- Dissolve in 1950 Β΅L of DMSO.
- Divide into 0.2 mL portions
- Store at -20 Β°C.
- Preparation of 250 mM stock solution (dissolved in DMSO):
- Β» O2k Manual Titrations: MiPNet09.12 O2k-Titrations
- Titration volume: 8 Β΅L
- Final conc. in 2 mL O2k chamber: 1 mM
