SUIT protocols have been developed for the first time on the basis of high-resolution respirometry, since their application depends critically on long-term stability of the polarographic oxygen sensor (experiments may be extended over 1 to 2 hours), highly diluted sample to avoid fast oxygen depletion in the closed chamber, and the possibility for re-oxygenations to extend measurement of oxygen flux. The mitochondrial preparation must be stable over such experimental periods of time, hence specific mitochondrial respiration media (MiR05: Gnaiger_2000_MitoInTheCold; MiR06) have been developed to maintain respiratory capacity during application of mitochondrial SUIT protocols at physiological temperature (Gnaiger_2007_MitoPathways).
